基质金属蛋白酶13和转化生长因子β1 mRNA及蛋白在骨性关节炎中的表达 被引量:3

Expression of MMP-13 mRNA,TGF-β1 mRNA and proteins in osteoarthritis

摘要 目的:探讨骨性关节炎(OA)中金属蛋白酶-13(MMP-13),转化生长因子-β1(TGF-β1)mRNA和蛋白表达的意义.方法:采用免疫组织化学技术,对60例OA病例及20例正常对照组进行了MMP-13及TGF-β1蛋白表达的检测;通过原位杂交技术,对30例OA及10例正常对照进行了MMP-13及TGF-β1在mRNA水平表达的检测.结果:MMP-13mRNA和蛋白在OA组阳性表达的积分光密度(IOD)值均显著高于正常对照组(P<0.01),OA中两者的表达有显著正相关性(P<0.01);TGF-β1mRNA和蛋白在OA组阳性表达的IOD值均显著高于正常对照组(P<0.01),OA中两者的表达亦有显著正相关性(P<0.01);MMP-13mRNA,TGF-β1mRNA及相应蛋白在OA中的表达均呈负相关(P<0.05或P<0.01).结论:OA中TGF-β1高表达通过下调关节软骨与滑膜细胞中MMP-13的表达,对OA关节软骨起保护作用. AIM:To investigate the significance of expression of MMP-13,TGF-β1 mRNA and corresponding proteins in Osteoarthritis.METHODS:Immunohistochemical(IHC)analysis was used to determine the distribution of MMP-13 and TGF-β1 protein in chondrocytes and synoviocytes of osteoarthritis from 60 patients with osteoarthritis group and 20 patients with normal control group.Expression of MMP-13 and TGF-β1 mRNA in chondric and synovial tissues was detected in 30 cases with osteoarthritis group and 10 cases with normal control group by in situ hybridization(ISH).RESULTS:IOD of MMP-13 mRNA and protein in OA was significantly higher than normal control group(P〈0.01),and there was significant difference between IOD of TGF-β1 mRNA and protein in OA and control group(P〈0.01).There were positive correlations both between mRNA and corresponding protein in OA(r=0.956,0.941,P〈0.01).There were also significantly negative correlations both between IOD of MMP-13 and TGF-β1 in OA detected by ISH and IHC(r=-0.385,P〈0.05;r=-0.603,P〈0.01),respectively.CONCLUSION:The higher expression of TGF-β1 may be protectarticular cartilage by downregulation the expression of MMP-13 of chondrocytes and synoviocytes in OA.
