摘要
本文为了研究人B因子(BF)等位基因变异型间是否存在抗原决定簇的差异,用两株针对不同抗原决定簇的抗人BF单克隆抗体(A2、C_9)对纯化的FF型和S_A、S_B亚型的BF进行等电聚焦,免疫印渍,结果发现C_9与FF、S_A S_B均呈阳性反应;A_2与S_B、FF呈阳性反应,但却不与S_A反应。从而说明在FF与S_A间,S_A与S_B间有抗原决定簇的差异,该抗原决定簇与A2相关。
In attempt to study the variation of antigenic determinants among some complement factor B allelic variants BFF type and BFSA, BFSB subtypes were investiged with two mouse anti-BF monoclonal antibodies (McAbs) :A2, C9. The two McAbs have been proved specific for different antigenic determinants in our laboratory.In this study we use A2, C9 as probes of antigenic determinants of BF allelic variants. Immuno-blotting of BFF, BFSAand BFSB which were puried subjected to isoeletric focusing revealed that C9 reacted with the three phenotypes, while A2 reacted with BFF and BFSB but did not bind to BFSA. That A2 did not react with BFSA demonstrates that there is one antigenic determinant (related to A2) variation between BFF and BFSA and between BFSA and BFSB.Therefore, we can infer that the substitution of local amino acids caused by point mutation and silent mutation which underlie the basis of BF polymorphism can cause the change of an antigenic determinant.
