摘要
目的:观察8-氧鸟嘌呤脱氧核苷三磷酸酶同源蛋白2(MTH2)及8-氧鸟嘌呤脱氧核苷(8-oxodG)在阿尔茨海默病(AD)患者海马组织标本中的表达变化,探讨在AD患者海马中二者表达的相关性。方法:选用临床及病理均诊断为AD的尸检海马组织石蜡切片作为实验组,年龄匹配且临床及病理诊断均排除AD的尸检海马组织石蜡切片作为对照组,每组各3例,用免疫组化的方法分别检测人脑海马中8-oxo-dG的含量及MTH2蛋白的表达。结果:免疫组化定量结果显示,在人脑海马组织CA1及CA3区,AD患者8-oxo-dG的表达显著高于年龄匹配对照组,而AD患者MTH2的表达显著低于年龄匹配的对照组。结论:在人脑海马组织中,DNA氧化产物8-oxo-dG含量在AD患者比年龄匹配对照组有显著提高,抗氧化修复蛋白MTH2的表达量却显著降低,提示人脑海马组织中MTH2蛋白表达量的减少及8-oxo-dG含量的增加与AD的发病过程相关。
Objective:To investigate the changes of 8-oxo-2'-deoxyguanosine (8-oxo-dG)amount and the expression of Mutt homolog 2(MTH2)in the hippocampi of Alzheimer's disease(AD)patients.Methods:The'aherations of 8-oxodG and MTH2 expression were analysed in hippocampi from autopsy specimen from three AD patients and three age-matched controls by means of immunohistochemistry using specific antibodies.Results: Semiquantitative immunohistochemical results indicated,in the CA1 and CA3 regions of hippocampus of AD patients,the contents of 8-oxo-dG were significantly higher than those in control subjects,and the expressions of MTH2 were significantly lower than those in control subjects.Conclusion:The increase of 8-oxo-dG and the decrease of the expression of MTH2 presented in a synchronizing style was observed in the hippocampus of AD patients.The result suggested that the ongoing accumulation of 8-oxo-dG and the difficiency of nucleotide repair enzyme MTH2 might be relevant with the pathogenesis of AD.
