摘要
白僵蚕体内所含的草酸铵为抗惊厥的有效药用成分。采用响应面法优化了白僵蚕草酸铵的提取工艺条件。在单因素试验的基础上,选择提取温度、提取时间、原料质量浓度3个因素进行中心组合试验,建立了草酸铵得率的二次回归方程。此模型拟合性好,通过响应面分析以及岭嵴分析得到了优化组合条件。试验结果表明,提取温度及原料质量浓度对草酸铵得率都有显著影响,当提取工艺条件为温度85℃,提取时间65 min,原料质量浓度43 g/L时,草酸铵得率达到最大值。此条件下草酸铵得率验证值为3.25%,与预测值3.29%基本一致。
Ammonium oxalate extracted from the white bodies of Bombyx Batryticatus is an effective medicinal ingredient of anticonvulsants.Response surface methodology(RSM) was used for optimizing the extraction technology of ammonium oxalate from Bombyx Batryticatus.Based on single-factor experiments,three levels of factors including extraction temperature,extraction time and material mass concentration were selected to conduct the central composite experiment.As a result,a quadric regression equation was established for the yield of ammonium oxalate.The equation had good simulation performance,from which an optimum combination was obtained from response surface analysis and ridge analysis on it.Experimental results showed that extraction temperature and material mass concentration affected the yield of ammonium oxalate remarkably.The optimal conditions for the extraction were 85 ℃,65 min and 43 g/L material mass concentration,under which the yield of oxalate ammonium was the highest.Under the optimal extraction conditions,the practical yield of ammonium oxalate was 3.25%,being very close to the predicted value 3.29%.
