感染水稻黑条矮缩病毒的玉米组织细胞的变化 被引量:1

摘要 在防虫网室内,人工接种水稻黑条矮缩病毒(Rice black-streaked dwarffijivirus,RBSDV)于3叶1心期至5叶1心郑单958玉米苗上。接种10 d后,每天调查症状,抽雄期测量植株上部幼嫩茎节长度;对接种病株和未接种健株的叶片进行横切;对茎节组织进行横切和纵切,荧光染色后,显微镜下观察横切片的细胞数量、形态变化,测量茎节纵切片的细胞长度。结果显示:与未接种的健康对照相比,受侵染的玉米叶片和茎节组织的韧皮部细胞畸形、数量剧增,且排列紊乱。感病植株的节间长度缩短,细胞伸长受阻。本研究通过组织细胞形态观察,明确了RBSDV侵染玉米后,所致内部组织细胞形态的变化和外部植株矮化、叶背蜡泪状突起等典型症状的相关性。 Artificial inoculation of Rice black-streaked dwarf fijivirus(RBSDV) on maize through viral vector,Laodelphax striatellus Fallen,was carried out in screen house.Symptoms were investigated after inoculation through the whole growth period and the younger stem was measured in the tasseling stage.The histological observation of the cross and longitudinal sections under fluorescence microscope of leaves and stems revealed that the phloem cells from diseased leaves and stems remarkably increased and the morphology was abnormal and disorganized compared with the healthy ones.Longitudinal section observation of last third internode from the top of maize stem indicated that the cell expansion was inhibited and the cell length of the diseased stems was shorter than the healthy ones.It was concluded that symptoms of gall formation(enations) in the abaxial epidermis of leaves correlated to the phloem cell increase,abnormal and disorganized morphology;stunted plants associated with the inhibition of stem cell expansion.
