摘要
利用抗坏血酸液相还原法制备适用于免疫检测用的胶体金纳米颗粒,并采用紫外/可见光分光光度计对胶体金颗粒的光学吸收特性进行表征。通过对实验中抗坏血酸的加入浓度、加热程度以及稳定剂作用的探讨,发现当氧化剂HAuCl4的浓度为1%时,反应体系中抗坏血酸浓度控制在1%,制备得到的胶体金颗粒较小且有较好的均一性;良好的加热并适量添加PEG能提高胶体金的质量。实验还发现胶体金溶液的最大吸收峰和其粒径之间存在一定的线性关系,其线性度达到0·9768,线性回归方程为Y=2·834X+474·16。
Ascorbic acid,as a reducing agent was applied to prepare the colloidal gold immunossay nanoparticles and the optical absorption properties of colloidal gold particles were measured by UV/visible light spectrophotometer.The effects of ascorbic acid concentration,the degree of heating and stabilizer PEG on colloidal gold particle were investigated.The results showed that at the condition of 1% ascorbic acid in 1% HAuCI4 reaction system,the particle diameter and homogeneity of the colloidal gold particles were better.The favorable heating and the appropriate addition of PEG can improve colloidal gold particles quality.The linear regression relationship (Y = 2.834X +474.16) existed between colloidal gold average particle size and wavelength of maximum absorption peak.
