摘要
在印刷电极表面涂覆一层壳聚糖(Chit)膜,在膜上同时固定四羧基酞菁铁(Ⅲ)(FePc)和HRP酶标黄曲霉毒素B1抗体(HRP-Ab-AFB1)包被纳米金,制备了可用于小麦粉中黄曲霉素B1(AFB1)快速检测的新型安培免疫传感器(SPCE|Chit/FePc/Au/HRP-Ab-AFB1)。FePc对H2O2的还原具有催化作用,可作为HRP酶与电极之间电子传递媒介体。当该传感器在含AFB1样品的30℃溶液中温育8 min后,AFB1与Ab-AFB1的免疫结合导致HRP的活性中心与FePc之间的电子传递被部分阻碍,使HRP对H2O2电催化氧化电流Io降低。ΔIo与AFB1浓度在1.0-200μg/L成线性关系,标准样品的组内和组间变异系数〈3.5%;回收率97%-104%,检测限为0.3μg/L。该传感器检测AFB1温育时间短,一步测定免分离,为小麦粉中的AFB1现场分析提供了新技术。
A novel amperometric immunosensor for rapid determination of aflatoxin B1(AFB1) in wheat flour based on AFB1 antibody coated nano gold particles modified electrode is proposed.The immunosensor is prepared by co-immobilizing HRP labeled monoclonal antibody of AFB1(HRP-AFB1),Fe(Ⅲ) Phthlocyanine on the surface of a nano Au collide-modified SPCE electrode by Christon membrane(SPCE| Chit/FePc/Au/HRP-Ab-AFB1).FePc can catalyze the reduction of H2O2 and act as electron mediator between the electrode and HRP during determination. After the immunosensor is incubated with AFB1 solution at 30℃ for 8 min, the access of activity center of the HRP to electrode is partly inhibited, which leads to a linear decrease of catalytic efficiency of the HRP to the oxidation of immobilized thionine by H2O2 in AFB1 concentration at 1.0 to 200 μg/L and the detection limit is 0. 3 μg/L. Being successfully utilized for determination of AFB1 antigen in extraction samples of wheat flour, the immunosensor shows a better accuracy and more rapidly. This method would be valuable for screen determination of AFB1 in flours.
