摘要
采用ISSR标记技术对40个姬菇杂交菌株及2个亲本菌株进行分析,使用NTSYSpe2.1生物软件对42个供试菌株进行聚类分析,构建系统树。试验筛选出了11个扩增谱带清晰、多态性好的ISSR引物,共扩增出81条清晰易辨的多态性谱带。聚类分析结果表明,42个供试菌株遗传相似系数变异范围为0.4286~0.8537,在相似系数0.6680时,42个菌株聚为7类,两亲本各聚一类,杂交菌株与亲本菌株遗传差异较大。
Inter-simple sequence repeat (1SSR) was used to determine the forty hybrid strains and their parents strains ( J1-2, J2-1 ). Genetic similarity coemcient and the dendrogram were obtained by NTSYS-pc( Version 2.10e) software. 11 primers which were screened from 75 1S- SR primers could be used to amplify the genomes of all these 42 test strains, and to gain the clear and stable fingerprint maps with strong polymorphism. The result showed that the total 116 bands were gained, and the similarity coefficients ranged from 0.4286 to 0. 8537. When the similarity coefficient was 0. 6680, forty-two strains separated in 7 glades were different from their diverse parents.
