摘要
将Epstein-Bar病毒(EBV)膜抗原(MA)BLLF1基因,插入含有CMV启动子的真核表达载体pcDNA3下游BamHI位点,构建成真核表达质粒pcDNA3-MA。将纯化的DNA注射Balb/c小鼠股四头肌。经免疫的动物产生抗EB病毒MA特异性的抗体和中和抗体,依赖抗体细胞介导的细胞毒作用(ADCC),特异性T淋巴细胞增生性反应及细胞毒性T淋巴细胞(CTL)杀伤作用。基因免疫与基因-蛋白联合免疫效果相似。不同启动子控制下的MA基因,诱发小鼠免疫应答无明显差异。
A recombinant plasmid DNA,pcDNA3-MA,was constructed.2.7kb fragment of Epstein-Barr virus(EBV)membrane antigen(MA)BLLF1 gene in which the anchor sequence was removed was inserted into pcDNA3 vector under the control of cytomegalovirus(CMV)promoter.Purified pcDNA3-MA and pMDⅢGPTR were injected into quadriceps muscles of Balb/c mice separately.The results showed that intramuscular inoculation of recombinant plasmid DNA generated in mice the specific antibody to EBV MA protein, neutralizing antibody and ADCC activity.Most importantly,the lymphoproliferative responses and cytotoxic T-lymphocyte activity were detected.The immune responses to gene alone,or to gene and protein combination were similar.Also there was no difference in immune responses between two plasmid DNA containing MA gene under control of SV40 or CMV promoter.Cell mediated immunity is an important feature of gene immunization.Thus,the DNA vaccination may become an effective method for preventing infectious diseases.
