摘要
目的:观察丹参酮ⅡA对兔动脉粥样硬化斑块胶原含量的影响,并对其机制进行初步探讨。方法:日本大耳兔随机分为对照组和丹参酮ⅡA(7、20、60mg/kg)4组,高胆固醇饮食10周建立兔动脉粥样硬化(atherosc lerosis,AS)病变模型后药物干预4周,取主动脉内膜组织分别采用W oessner方法、免疫组织化学方法和realtim e RT-PCR法检测斑块组织中胶原含量,MMP-1蛋白,IL-10、IL-1 mRNA表达,W estern-b lot方法检测斑块组织中PPAR-γ蛋白表达。结果:丹参酮ⅡA低剂量能明显提高斑块中胶原蛋白含量、PPAR-γ蛋白和IL-10 mRNA表达,明显降低斑块组织中MMP-1和IL-1 mRNA的表达;中、高剂量组能显著提高斑块中胶原蛋白含量、PPAR-γ蛋白和IL-10 mRNA表达,显著降低斑块组织中MMP-1和IL-1 mRNA的表达。结论:丹参酮ⅡA能提高动脉粥样硬化斑块中胶原蛋白含量,其机制可能与抑制炎症反应,降低MMP-1表达有关。
Aim: The instability of atherosclerosis plaque was the critical factor induced the heart events,and valid remedies for this were deficient at present.Rabbit atherosclerotic models were set up to study effects of Tanshinone ⅡA on stability of atherosclerotic plaques and initial investigation for mechanism.Method: Rabbit atherosclerotic models were set up by hypercholestermia diet.Contents of matrix metalloproteinase-1 and collagen in plaque were detected by immunohistochemical and woessner method,respectively.contents of metalloproteinase-1 protein were measured by Image Pro Plus 4.5.Expression levels of interleukin 6,interleukin-10 mRNA and peroxisome Proliferator-activated receptor gamma protein were determined with real-time PCR and western blot method,respectively.Result: In Tanshinone ⅡA groups,collagen,Proliferator-activated receptor gamma protein and interleukin 10 mRNA was elevated markedly,interleukin 6 mRNA and MMP-1 protein found in Tanshinone ⅡA groups were decreased.Conclusion: Tanshinone ⅡA could enhance stability of rabbit atherosclerotic plaque.This maybe be concerned with its anti-inflammatory action.
