摘要
目的:研究丹参的三种单体丹酚酸A、紫草酸和迷迭香酸干预心肌缺血的可能环节。方法:体外培养大鼠原代心肌细胞;三种单体与细胞共孵育24小时,观察细胞活力的变化,确定药物作用安全范围;复制心肌细胞过氧化氢及缺氧复氧损伤的模型,使用三种单体进行干预,分别观察细胞活力和LDH漏出的改变的改变,评价三种单体的抗氧化和抗缺氧作用。结果:在100 mg/L以下的丹酚酸A和500 mg/L以下的紫草酸、迷迭香酸均不降低心肌细胞的活力;25~100 mg/L的三种单体均能显著地抑制过氧化引起的细胞活力的下降;12.5~100 mg/L的丹酚酸A和50~100 mg/L的迷迭香酸还可以显著抑制缺氧复氧导致的细胞LDH漏出。结论:丹酚酸A、紫草酸和迷迭香酸对心肌细胞具有较高的安全性,同时具有显著的抗心肌细胞氧化损伤的能力;丹酚酸A和迷迭香酸还具有显著的抗心肌细胞缺氧损伤的能力。这三个单体的对心肌细胞的直接保护作用,可能是丹参治疗心肌缺血的机制之一,它们可能是丹参药理活性的物质基础。
Objective:To study the possibly interventional parts of three components of salviae miltiorrhizae radix,salvianolic acid A,lithospermic acid and rosmarinci acid,in myocardium ischemia,elucidate the therapeutic mechanisms and marterial basis of salviae miltiorrhizae radix on myocardium ischemia.Method: Neonatal rat cardiomyocytes were cultured in vitro;the three compounds were incubated with cardiomyocytes for 24 hours,and the cell viability was measured to determine the safe concentrations of them;the hydrogen peroxide and an anoxia-reoxia injury models were duplicated in cardiomyocytes,and the three compounds were treated to the cell,the cell viability and LDH leakage was measured to value to the effects of the compounds on the two models,respectively.Result: Salvianolic acid A,lithospermic acid and rosmarinci acid did not sinificantly decrease the cell viability,at most up to 100,500 and 500 mg/L,respectively;all of the three compounds significantly inhibited the decrease of cell viability induced with hydrogen peroxide,at concentrations of 25~100 mg/L;Salvianolic acid A and rosmarinci acid significantly inhibited LDH leakage induced by anoxia-reoxia injury,at concentrations of 12.5~100 and 50~100 mg/L,respectively.Conclusion: Salvianolic acid A,lithospermic acid and rosmarinci acid have high safeties and significant anti-peroxidation effects on cardiomyocytes;salvianolic acid A and rosmarinci acid have significant anti-anoxia effects on cardiomyocytes;the direct protective effects on cardiomyocytes of the three compounds may be one of the therapeutic mechanisms of salviae miltiorrhizae radix on myocardium ischemia and the three compounds may be parts of the marterial basis of the pharmacological activity of salviae miltiorrhizae radix.
