不同激素配比对紫花苜蓿不同外植体愈伤组织诱导的影响 被引量:5

摘要 [目的]为紫花苜蓿外植体培养筛选最佳激素配比培养基。[方法]分别以盛世无菌苗的下胚轴、子叶、叶片、叶柄、根为外植体,在MS附加不同激素配比的培养基上诱导愈伤组织,研究不同处理对愈伤组织诱导率的影响。[结果]5种外植体均能诱导出愈伤组织,其中下胚轴的愈伤组织诱导率最高(达87.5%),且愈伤组织长势良好,其次为子叶>叶柄>叶片>根;不同外植体愈伤组织的形成时间为下胚轴>子叶>叶柄和根>叶片;2,4-D诱导愈伤组织的效果优于NAA。[结论]2,4-D、NAA、6-BA和KT配合使用的培养基上形成的愈伤组织浅黄带绿,表面突起,松软,带簇状芽或单芽或形成胚状体。 [ Objective ] The aim was to select the medium with the optimum hormone combination for explant culture of alfalfa. [ Method ] With hypocotyl, cotyledon, leaf, petiole and root of "shengshi" as the explants, the callus was induced on MS medium with different hormone combination. The effects of different treatments on induction rate of callus were studied. [ Result ] The callus could be induced from 5 kinds of explants, in which the induction rate of callus from hypocotyl was the highest ( reaching 87.5% ) and the callus all grew well, and the following was that of cotyledon 〉 petiole 〉 leaf 〉 root. The formation time of callus from different explants showed as hypocotyl 〉 cotyledon 〉 petiole and root 〉 leaf. The induction effect of callus with 2,g-D was better than that with NAA. [ Conclusion ] The callus on the medium with 2,4-D, NAA, 6-BA and KT was light yellow-green with processes on their surface, and their structure was fluffy with cluster bud or single bud or embryoid.
