摘要 [目的]提高枸杞花药培养的诱导率。[方法]以宁杞1号、大麻叶、0207、蒙杞1号为材料,以B5为基本培养基对其花药进行离体培养,考察不同激素组合、活性炭、甘露醇预处理对愈伤组织形成的影响。[结果]宁杞1号是比较容易诱导的基因型,0207是较难诱导的基因型;2,4-D对愈伤组织诱导具有明显促进作用;培养基中NAA为0.1 mg/L、6-BA为1.0 mg/L时,胚状体诱导率达3.5%;添加活性炭可明显提高胚状体的诱导率,培养基中活性炭为0.1 mg/L、NAA为0.1 mg/L、6-BA为1.0 mg/L时,胚状体诱导率可提高6倍;一定浓度的甘露醇预处理可提高花药愈伤组织的诱导率。[结论]材料基因型和培养基激素配比是影响枸杞花药培养的重要因素。 [ Objective ] The aim was to increase the induction rate in anther culture of Lycium bararum L.. [ Method ] With Ningqi 1, Damaye, 0207 and Mengqi 1 as the materials and B5 as the basic medium. The anther of L. bararum was cultured in vitro, and the effects of different hormone combinations, activated carbon and pretreatment with mannitol on formation of callus were studied. [ Result] Ningqi 1 was the genotype that was easy to induce and 0207 was the genotype that was difficult to induce. 2,4-D had obvious promotion on callus induction. When NAA was 0. 1 mg/L and 6-BA was 1.0 mg/L in medium, the induction rate of embryoid reached 3.5%. Activated carbon could obviously increase the induction rate of embryoid. When activated carbon was 0. 1 mg/L in medium with NAA of 0. 1 mg/L and 6-BA of 1.0 mg/L, the induction rate of embryoid was increased by 6 times. Pretreatment with mannitol at a cetern concn, could increase the induction rate of callus. [ Conclusion] The genotype of materials and the hormone combinations in medium were the main factors that affect the anther culture of L. bararum.
