摘要 目的:近年来A型肉毒毒素(BTX-A)在疼痛治疗中的作用已成为新的热点,文中研究腹腔注射BTX-A在大鼠内脏性疼痛中的镇痛作用,以及对肠壁乙酰胆碱酯酶(AChE)、P物质(SP)表达的影响。方法:72只成年雄性Wistar大鼠随机分为4组,A组腹腔注射等渗盐水2 ml,B、C、D组分别腹腔注射含2、4、6 UBTX-A的等渗盐水2ml,注射后1、4、8周时,各组随机抽取6只大鼠进行扭体实验疼痛评分,留取肠道标本行AChE、SP免疫组化检测。结果:1周时,大鼠扭体反应疼痛积分:C、D组大鼠均明显少于A组(P<0.05),B、C、D组肠壁肌层AChE表达及C、D组肠壁SP表达均小于A组(P<0.05)。4周时,B、C、D组疼痛积分明显少于A组(P<0.05);B,C,D组肠壁AChE、SP表达均明显小于A组(P<0.05)。8周时,C组疼痛积分与A组相比减少;各组肠壁AChE表达仍小于A组(P<0.05)。结论:腹腔注射BTX-A能减少乙酸所致大鼠内脏性疼痛的疼痛积分,并能减少肠壁AChE、SP的表达。 Objective: In recent years,botulinum toxin type A(BTX-A) has been investigated for the treatment of pain.This experiment was to investigate the antinociceptive effect of BTX-A intraperitoneal injection on visceral pain of rats and its effect on intestinal AChE and SP expression.Methods: 72 male adult Wistar rats were divided into four groups: BTX-A(2U,4U,and 6U) or vehicle(2ml) was injected intraperitoneally in B,C,D and A group.6 Rats of every group were challenged with acetic acid intraperitoneal injection after 1,4 and 8 weeks respectively.After abdominal writhing behaviors were monitored,the intestinal samples were immunohistochemical stained for AChE and SP examination.Results: At the end of one week,writhing test scores of rats in group C and D were decreased significantly compared with group A(P〈0.05).The expression of AChE in intestinal muscular layer of all BTX-A pretreated groups,and SP in intestinal walls of group C,D showed a significant decrease(P〈0.05).Four weeks later,the writhing test scores and the expression of AChE and SP of BTX-A pretreated groups all showed significant difference compared with those of group A(P〈0.05).After 8 weeks injection,the writhing test scores of group C and the expression of AChE in intestinal wall of all BTX-A treated groups were still lower than that of group A(P〈0.05).Conclusion: Injection of BTX-A intraperitoneally significantly reduces acetic acid-induced nociceptive behaviors and inhibits the expression of AchE and SP in intestinal walls of rats.
