摘要
目的:观察氯胺酮不同浓度在不同时间对离体Sprague-Dawley大鼠乳头肌收缩性能的影响。方法:10条乳头肌分别给予氯胺酮10、20、30、40μg/ml,观察等长张力峰值(PT),张力上升最大速度(dT/dtmax)、张力下降最大速度(-dT/dtmax)和心肌收缩成份缩短速度(Vce)在用药后不同时间的变化。结果:当氯胺酮的浓度大于20μg/ml,特别是达到40μg/ml时,PT、±(dT/dtmax)5min内即明显下降(P<0.05),10min时分别下降57.7%±3.2%,57.3%±4.1%和58.2%±3.8%,差异非常显著(P<0.01),15min即下降为零,有极显著性差异(P<0.001)。
Objective:To evaluate the effect of various concentration of ketamine at different time on myocardial contractility.Methods:Ten left ventricular papillary muscles in vitro were administrated respectively with 10、20、30、40μg/ml of ketamine.The contractility parameters of peak twitch tension(PT),maximum rate of contraction (dT/dt max ),maximum rate of relaxation ( dT/dt max ) and velocity of shortening of myocardial muscle contractility element (V ce ) were measured.The changes of myocardial contractility were observed at 2,5,10,15min after the virous concentrations.Results:Ketamine induced a slight change in PT,±dT/dt max ( P> 0 05) in the concentrations ranged from 10μg/ml to 30μg/ml.PT,dT/dt max and dT/dt max at 5 min decreased significantly by 57 7%±3 2%,57 3%±4 1% and 58 2%±3 8%,respectively in 40μg/ml of ketamine.The complete inhibition occured and lasted for 15min when concentration of ketamine was 40μg/ml.Conclusion:Cardiovascular depression observed with ketamine is related to its intrinsic myocardial depression.
