摘要
目的观察银杏叶提取物(ginkgobiobaextract,GBE)对大鼠肾间质成纤维细胞增殖能力的影响,及其对血管紧张素Ⅱ(AngⅡ)诱导肾间质成纤维细胞表达转化生长因子β1(TGF-β1)和结缔组织生长因子(CTGF)表达的抑制作用。方法取SD大鼠肾间质成纤维细胞进行体外培养并鉴定,以未处理细胞作为正常对照组(C),将AngII作为刺激因素,分别加入终浓度为0,25,50,100和200mg.L-1的GBE,采用MTT法检测GBE对各组细胞增殖能力的影响,RT-PCR检测各组细胞TGF-β1和CTGFmRNA表达。结果与正常对照组比较,AngⅡ刺激后使细胞增殖能力明显增强(P<0.05),AngⅡ的这种作用可被含GBE的培养液所抑制,且与GBE的浓度和培养时间有关;RT-PCR结果显示,AngⅡ组细胞TGF-β及CTGFmRNA表达量明显高于正常对照组(P<0.01),GBE则可以抑制AngⅡ诱导的TGF-β及CTGFmRNA表达量的上调(P<0.05,P<0.01)。结论银杏叶提取物能抑制大鼠肾成纤维细胞增殖,并且下调AngⅡ诱导的TGF-β及CTGFmRNA的过表达,这可能是银杏叶提取物防治肾纤维化的作用机制之一。
Objective To observe the effects of ginkgobiobaextraet (GBE) on the proliferation of the renal interstitial fibroblasts of rats (RFB), and the effects of GBE on Angll-induced TGF-β1, CTGF mRNA expression. Melhods The rat renal fibroblasts were cultivated in vitro and identified, untreated cells were acted as normal control group, Ang Ⅱ as the stimulating factor, the cells were treated with GBE with concentrations of 0, 25 , 50,100 and 200 mg· L- 1, Cell proliferation was observed by MTT assay, the mRNA levels of TGF-β1 and CTGF were measured by RT-PCR. Results Compared With the control group, Ang II significantly increased the proliferation of RFB ( P 〈 0.05) ,it can be inhibiton by GBE;Ang Ⅱ can increased the expression of TGF-β1 and CTGF mRNA ( P 〈 0.01 ), GBE can attenuated Ang Ⅱ -induced TGF-β1 and CTGF mRNA overexpression ( P 〈 0.01, P 〈 0.05). Conclusion Ginkgob- iobaextract can depress RFB proliferation and down regulation the AnglI-induced TGF-β1 and CTGF mRNA overexpression, suggesting it may be one of mechanisms underlying GBE in prevention and therapy the renal interstitial fibrosis.
