Separation of Angiotensin-I-Converting Enzyme Inhibitory Peptides Derived from Jellyf ish with Ultraf iltration Technology

摘要 目的探讨分离提取海蜇水解液中血管紧张素转化酶(ACE)抑制肽的有效方法。方法采用超滤技术分离ACE抑制肽,用凝胶排阻色谱Sephadex G-25分析超滤前后肽相对分子质量的分布,用高效液相色谱法测定超滤前后肽ACE的抑制率。结果超滤可明显改变水解液中肽相对分子质量的分布,将ACE抑制率从35.05%提高到58.85%(测定浓度均为0.56 mg/mL)。结论超滤明显改变了肽相对分子质量的分布,截留了ACE抑制率较低的大分子肽,提高了滤过液ACE抑制率。但是,超滤的相对流速有待提高。 Objective To investigate the efficient method for the separation of angiotensin-I-converting enzyme (ACE) inhibitory peptides derived from the hydrolysate of jellyfish. Methods The ultrafiltration technology was adopted for the separation of ACE inhibitory peptides. The molecular weight distribution of filtered and unfiltered peptides was investigated by Gel Exclusion Chromatography (Sephadex G-25), and the ACE inhibition rate of filtered and unfiltered peptides was determined by HPLC. Results The ultrafiltration could change the molecular weight distribution of peptides markedly and raise the ACE inhibition rate from 35.05 % to 58.85 % at the concentration of 0.56 mg/mL. Conelusion The peptides with lower ACE inhibition rate can be trapped by Ultrafiltration. Correspondingly, the ACE inhibition rate of the filtrate increases, but the relative flow rate should be improved.
