摘要
目的模拟高原条件下建立SD大鼠牙周炎动物模型,检测其龈沟液中C-反应蛋白(C-reactive protein,CRP)变化,以探索高原牙周炎发病机制。方法选取60只SD大鼠,雌雄各半,采用随机数字表法分为6组:平原对照组(A组)、平原牙周炎组(B组)、高原对照组(C组)、高原牙周炎组(D组)、平原牙周炎模型判定组(E组)、高原牙周炎模型判定组(F组),每组10只。B组和E组,D组和F组分别建立平原牙周炎和高原牙周炎动物模型,8周后E、F组判定造模成功,采用ELISA法检测剩余4组大鼠龈沟液中CRP浓度变化。结果A、B、C、D组龈沟液中CRP浓度分别为(6.33±1.58)、(13.47±1.98)、(7.76±1.84)、(16.26±2.14)μg/ml,B组与A组,D组与C组,C组与A组,D组与B组比较差异均有统计学意义(P<0.05),其中以D组升高最为显著。结论高原牙周炎组大鼠龈沟液中的CRP水平高于平原牙周炎组,提示高原环境下牙周炎的病变程度更严重。
Objective To establish a SD rat model of periodontitis in a simulated plateau-condition and investigate the change of C-reactive protein(CRP) concentrations in the gingival crevicular fluid(GCF) so as to explore the pathogenesis of plateau periodontitis.Methods Totally 60 SD rats(30 males and 30 females) were randomly divided into 6 groups,group A: plain control group;group B: plain periodontitis group;group C: plateau control group;group D: plateau periodontitis group;group E: plain periodontitis model assessment group;group F: plateau periodontitis model assessment group,and each group had 10 rats.Periodontitis was produced with the conventional method of Fischer by silk ligament suture around the cervical portion of first molars and fed with Keyes's diet.The models of plain periodontitis and plateau periodontitis(5 000 m) were established in group B and E,and group D and F respectively.Eight weeks later,group E and group F were killed and pathologically examined to make sure the model successfully established.ELISA was employed to determine the changes of CRP concentrations in GCF among remaining 4 groups.Results The CRP concentrations in GCF were(6.33±1.58),(13.47±1.98),(7.76±1.84) and(16.26±2.14) μg/ml in group A,B,C and D,respectively.There were statistic differences between group B and group A,group D and group C,group C and group A,group D and group B(P0.05).The increase of CRP concentration in group D was the most significant.Conclusion The CRP level in plateau periodontitis group is higher than that in plain periodontitis group,indicating that the severity of periodontitis is significantly higher in plateau.
