肝肽、脾肽对荷瘤裸鼠肿瘤细胞中Bcl-2、Bax表达的影响 被引量:1

THE EFFECTS OF ACBP-L AND ACBP-S ON EXPRESSION OF BCL-2 AND BAX IN GASTRIC CANCER-LOADED NUDE MICE

摘要 目的:观察两种抗癌生物活性肽对荷瘤裸鼠肿瘤细胞中Bcl-2、Bax表达的影响。方法:建立人胃癌裸鼠模型,裸鼠随机分为三组:对照组11只、脾肽实验组12只,肝肽实验组11只;分别尾静脉注射生理盐水0.2mL、脾肽0.2mL,肝肽0.2mL,隔日给药2wk后处死裸鼠,剥取肿瘤,以4%多聚甲醛固定,常规石蜡包埋,4μm切片。应用免疫组化法检测Bcl-2、Bax蛋白在实验组与对照组中的表达情况。结果:Bcl-2蛋白在对照组肿瘤中阳性表达较强,肝肽、脾肽组裸鼠肿瘤中Bcl-2蛋白的表达均比对照组表达弱,且有显著性差异(P<0.001);Bax蛋白在对照组肿瘤中阳性表达较弱,在肝肽组、脾肽组裸鼠肿瘤中Bax蛋白的表达均比对照组的表达强,有显著性差异(P<0.001)。结论:肝肽、脾肽具有较好的抑制荷胃癌裸鼠肿瘤生长的作用,其作用机制与影响Bcl-2及Bax的表达相关。 Objective:To explore the effects of the expression changes of anti - cancer bioacfive peptide - liver (ACBP - L) and anti - cancer bioactive peptide - spleen ( ACBP - S) on the expression of Bax and Bcl - 2 in gastric cancer - loaded nude mice. Methods: The nude mice xenografts model with gastric carcinoma cell were established, nude mice were divided into three groups randomly: 11 cases of nude mice as control group, 12 cases of nude mice as ACBP - L treatmend group and 11 cases of nude mice as ACBP - S treatment group. Three groups of nude mice were administrated into the tail vein of the mouse every day with 0.2mL physiologic saline, 0.2mL ACBP - L and ACBP - S respectively until two weeks. The mice were killed after one month , tumors were taken off and weighed. The tumors were decohesed and fixed with 4% paraformaldehyde, rutine paraffin embedded and sliced into 4μm. Then Bax and Bcl -2 expression were detected using immunohistochemistry SABC method between experimental and control groups. Results:The expression of Bax was relatively weaker in control group than those in experimental groups and which showed significant difference (P 〈0. 001 ). While the expression of Bcl -2 was stronger in control group than those in experiment groups and which showed significant difference (P 〈 0. 001 ). Conclusion: ACBP - S and ACBP - L have marked inhibiting effects upon the growth of BGC -823 cell in nude mice. The mechanism is probably related with its regulatory effects on Bcl -2 and Bax.
