摘要
目的:观察不同浓度的清痹合剂及清痹合剂+甲氨蝶呤(MTX)对大鼠佐剂性关节炎的影响及小鼠镇痛试验的观察。方法:1.皮下注射完全弗氏佐剂使大鼠关节致炎,复制RA大鼠模型。给药组分别以不同浓度的清痹合剂灌胃,观察大鼠的一般情况,右后足踝关节直径及足跖厚度变化;并对右后足踝关节滑膜进行病理观察。2.给各组小鼠灌以不同浓度的清痹合剂1h后,以0.6%醋酸生理盐水给小鼠按0.01ml/g体重腹腔注射,观察各组小鼠的扭体反应,记录20min内小鼠扭体次数。结果:清痹合剂高、中浓度对佐剂性关节炎肿胀有抑制作用(P<0.05,P<0.01),其中中浓度+MTX组对炎症抑制作用最为突出(P<0.01);清痹合剂与小剂量的MTX联合治疗,具有减轻滑膜充血、水肿、肉芽肿形成,抑制血管翳的形成。清痹合剂能减少醋酸致痛的小鼠扭体次数(P<0.05,P<0.01)。结论:清痹合剂对大鼠佐剂性关节炎有明显的消炎、镇痛作用,能很好协同小剂量MTX抑制滑膜血管翳的形成。
Objective：To observe the effect of different concentrations of Qingbi compound and Qingbi compound plus methotrexate（MTX）in rat adjuvant arthritis and the analgesic effect in mice.Methods：1.Rat adjuvant-induced joint inflammation was made by subcutaneous injection of complete Freund＇s adjuvant.Treatment groups were fed with different concentrations of Qingbi compound.Then the ankle joints＇ diameter and the paw thickness of the right hinds and the general situation were observed.Meanwhile,the synovial pathological observation of the right rear ankle joints was performed.2.The mice in each group were given different concentrations of Qingbi compound by intragastric administration.One hour later,0.6% acetic acid saline solution was used by intraperitoneal injection at 0.01ml /g.The reactions of the mice and the numbers of writhing within 20 minutes were observe d and recorded.Results：Qingbi compound of high and medium concentrations significantly inhibited the swelling of adjuvant-induced arthritis（P0.05,P0.01）,and the best effect was observed in medium concentration of Qingbi compound plus MTX（P0.01）.The combination therapy of Qingbi compound with low doses of MTX reduced synovial hyperemia,edema,granuloma formation and inhibited pannus formation as well as reduced writhing times of mice induced by acetic acid（P0.05,P0.01）.Conclusion：Qingbi compound has obvious anti-inflammatory,analgesic effect on adjuvant arthritis in rats,and can also inhibit the formation of synovial pannus with small dose of MTX.
