摘要
[目的]建立高效液相色谱-蒸发光散射法(HPLC-ELSD)测定枸骨叶中熊果酸的含量。[方法]采用phe-nomenexC18柱(5μm,4.6mm×250mm),流动相:甲醇-0.5%醋酸铵溶液(92:8);检测器:蒸发光散射检测器(Alltech ELSD 400)。[结果]在此色谱条件下,熊果酸与干扰峰完全达到基线分离(R>1.6)。枸骨叶熊果酸含量均在1.34%以上。熊果酸含量测定的线性范围为0.648~6.48μg(r=0.9998),平均加样回收率为97.1%,相对平均标准偏差(RSD)为1.7%。[结论]该法快速,简便,重现性好,可为《中国药典》中枸骨叶药材的质量标准提高提供参考。
[Objective] To establish HPLC-ELSD method for measuring the ursolic acid in leaves of Ilex cornuta. [Methods] The column of phenomenex C18(5μm,4.6mm×250mm) was used in this study and methanol-0.5% ammonium acetate (92:8) as the mobile phase. The detector was Alhech ELSD 400. [Results] Chromatographic separation of ursolic acid and the interfering peak was separated completely(R〉 1.6). The content of ursolic acid in leaves of Ilex comuta was more than 1.34% and in the ranges of 0.648-6.480μg(r=0.9998). The average recovery was 97.1% and RSD was 1.7%. [Conclusion] This method is rapid, easy to reproduce and can provide the basis to improve the quality of Ilex cornuta in the Chinese Pharmacopeia (ChP).
