摘要
试验研究了不同培养基对秦党叶片及茎段培养的效应。结果表明,在光照强度1600 lx,温度25℃的条件下,利用组织培养技术快速繁殖秦党时,适宜的外植体为幼苗的茎段。茎段培养时,诱导愈伤组织并促进分化的适宜培养基组分为:MS+2,4-D 0.2 mg/L+蔗糖20 g/L+琼脂8 g/L。仅促进芽生长适宜的培养基组分为:MS+BA2.0 mg/L+IAA1.0 mg/L+蔗糖20 g/L+琼脂8 g/L。仅诱导愈伤组织适宜的培养基组分为:MS+BA1.0 mg/L+IAA1.0 mg/L+蔗糖20 g/L+琼脂8 g/L。
It had studied the effect of different culture medium to the culture of Codonopsis tsinlingensis' lamina and stem, and the results showed that when the intensity of illumination was 1600 lx and the temperature was 25℃, the suitable explant was the young stem when using tissue culture technology to breed Codonopsis tsinlingensis quickly. The culture medium that could inducement callus and accelerate differentiation was MS+2,4-D 0.2 mg/L+sucrose 20 g/L+agar 8 g/L.The culture medium that only could accelerate bud growth was MS+BA2.0 mg/L+IAA1.0 mg/L+sucrose 20 g/L+agar 8 g/L.The culture medium that only could inducement callus was MS+BA1.0 mg/L+IAA 1.0 mg/L+sucrose 20 g/L+agar 8 g/L.
