摘要
目的:通过研究心肌微环境因素对S180细胞的影响,探讨心肌内转移瘤罕见的机制。方法:原代培养新生Wistar大鼠心肌细胞,MTT法分析心肌细胞条件性上清液(CMCM)对S180细胞的体外抑制作用。以顺铂作为CMCM的阳性对照,以正常细胞鼠肾小球系膜细胞为阴性对照。采用胰酶消化法,热灭活法初步探讨心肌细胞培养液的理化性质。结果:S180细胞与CMCM共同培养后,细胞存活率明显下降,与DMEM相比差异均具有显著性(P<0.05),而对正常细胞存活率无明显影响(P>0.05)。DDP对S180细胞和正常细胞均具有显著性抑制作用(P<0.05)。心肌细胞培养液经胰酶消化后活性仍然存在,而经热灭活后活性消失。结论:新生大鼠心肌细胞上清液可选择性的抑制S180细胞的增殖;这种抑制作用与化疗药物顺铂作用机制不同。心肌微环境中可能存在某种抑瘤物质,是心肌转移瘤罕见性的主要机制。
Objective:To explore the effect of organic microenvironment of cardiac muscles on the carcinoma cell S180 ,and to investigate the mechanism of the rarity of metastases in cardiac muscles. Methods: Primary culture of newborn Wistar rat cardiac muscle cells was established, and the murine cardiac muscle conditioned medium(CMCM) was prepared to test its effect in vitro on S180 by MTT assay. Cisplatin was used as positive control for CMCM,DMEM was used as negative control. Murine benign renal cells - mesangial cells were used as negative control for S180 cells. The cardiac muscle cells conditioned medium( 1:1 ) was also tested boiling at 100℃ and treatment with trypsin. Resiuts: Proliferation of tumor cell lines of S180 was significantly restrained when cultured with CMCM, while the routine benign renal cells were not affected( P 〈 0.05 ). Conclusion: Cardiac muscle cells could selectively inhibite the proliferation of cancerous cells in vitro while benign cells are not affected.
