摘要
目的探讨山药多糖对卡介苗(BCG)与脂多糖(LPS)致小鼠免疫性肝损伤的保护作用。方法将昆明种小鼠分为对照组、模型组、山药多糖高、中、低剂量保护组,第1天,模型组与保护组均尾ivBCG,保护组每天分别按体重30、60、120mg·kg-1山药多糖ig一次,第12天末次ig2h,模型组与山药多糖高、中、低保护组经尾ivLPS。16h后取小鼠称重,于眼球取血,测定小鼠血清中ALT、AST活性;处死小鼠,计算肝指数和脾脏指数;制备肝匀浆,测定肝组织中MDA、GSH含量及SOD和GSH-Px活性。结果与模型组比较山药多糖各剂量可降低小鼠肝指数、脾脏指数及降低血清ALT、AST活性(P<0.05);降低MDA的含量,增加GSH含量和GSH-Px活性。结论山药多糖对BCG和LPS诱导的小鼠免疫性肝损伤有保护作用。
OBJECTIVE To investigate the protective effect of the yam polysaccharide on the BCG and LPS - induced immunological liver injury in mice. METHODS Kunming mice were divided into control group, model group, low - dosage, intermidiate - dosage and high - dosage yarn polysaccharide groups. On the first day, the model group and the protection groups were injected BCG from tail veil and then the protection groups were administered with yam polysaccharide by gastric peffusion 30,60,120 mg·kg^-1. At the end of 12 days ,2 hr after gavage, the model group, high -dosage and intermidiate -dose yam polysaceharide groups were injected with LPS via the tail vein. 16 hr later, mice were weighed. The serum ALT and AST activity were determined and the mice were killed to calcu- late liver somatic index and spleen index. Liver homogenate were prepared and liver tissue MDA, GSH content and SOD and GSH - Px activity were tested. RESULTS The yam polysaccharide groups reduced the somatic index, spleen index and decreased serum ALT, AST activity of mouse liver, compared with the model group ( P 〈 0.05 ) and also decreased MDA content, increased GSH content and GSH - Px activity, compared with the model group (P 〈 0.05). CONCLUSION Chinese yarn polysaccharlde protected the BCG and LPS -induced immunological liver injury in mice.
