创伤弧菌脓毒症大鼠肺组织NF-κB和IL-18的基因表达以及药物的干预效应 被引量:4

Gene Expression of NF-κB and IL-18 in Lungs of Septic Rats with Vibrio vulnificus and Its Intervention Effects from Antibacterial Agents

摘要 目的探讨创伤弧菌脓毒症大鼠肺组织NF-κB和IL-18的基因表达以及头孢哌酮和左氧氟沙星联合抗炎治疗对其的干预效应。方法SD大鼠随机分为正常对照组、创伤弧菌脓毒症组(AA组)、创伤弧菌脓毒症联合抗炎治疗干预组(VV组),取各组部分肺组织,采用逆转录-聚合酶链反应方法测定各组NF-κBmRNA和IL-18 mRNA的表达量,并采用SPSS10.0软件进行统计学分析。结果VV组各组大鼠肺组织NF-κBmRNA和IL-18 mRNA表达量均明显高于NC组(P<0.05);AA组大鼠肺组织NF-κBmRNA、IL-18 mRNA表达量呈降低趋势,与相应时间点的VV组相比均明显降低(P<0.05)。结论创伤弧菌脓毒症大鼠肺组织NF-κB mRNA、IL-18 mRNA在脓毒症早期即增高,及早使用头孢哌酮和左氧氟沙星可明显减低其表达水平,从而有利于抑制炎症反应。 OBJECTIVE To study the gene expression of nuclear factor-kappaB(NF-kB) mRNA and IL-18 mRNA in the lungs of septic rats with Vibrio vulnificus and find out its intervention effects of cefoperazone sodium combined with levofloxacin. METHODS SD rats were randomly divided into normal comparison group (NC group), V. vulnificus sepsis model groups (VV groups) and drug intervention model groups(AA groups). Some lung tissue was obtained from rats. The concentration of NF-kB mRNA and IL-18 mRNA was measured by reverse transcription-polymerase chain reaction, and analyzed by SPSS10.0. RESULTS Compared with the NC group, the expression of NF-kB mRNA and IL-18 mRNA in the VV groups were increased significantly (P〈0.05). The expression of NF-kB mRNA and IL-18 mRNA in the AA groups were reduced gradually, compared with the VV groups, the expression was diminished greatly in the AA groups at the same time after being injected VV (P〈0.05). CONCLUSIONS The expression of NF-kB mRNA and IL-18 mRNA in the lungs of the rats reach the peak in the early period of VV sepsis. The treatment with cefoperazone sodium and Levofloxacin should be used as early as possible, because they reduce the expression, thereby inhibiting the inflammatory response in VV sepsis.
