摘要
[目的]比较具干细胞特性食管癌耐放射细胞株与其亲本基因表达水平的差异。[方法]Agilent4×44K人全基因组芯片检测具干细胞特性食管癌耐放射细胞株与其亲本细胞,GeneSpring GX 10.0.2数据分析挑选差异表达基因,进一步对差异表达基因进行GO分析和干细胞密切相关的信号通路分析,Real-time PCR验证芯片结果。[结果]两组细胞有明显不同的表达谱,差异表达基因多达2246个(fold change cut-off≥2)。在分子功能中,信号感应激活最高,占75.11%;在生物过程中,细胞进程、生物调节分别占35.94%和25.8%;在细胞组分方面,细胞、胞内区、跨膜区的变化比较显著。ALDH1基因表达发生了改变,其中ALDH1A3最显著。Hedgehog通路,PTCH1下调,而GLI2上调。BMP-TGFβ通路信号转导中,MMP3、MMP9上调,FOXL1、BMP4、TGFB2、TGFB3下调。Notch通路相关信号基因均表现为下调。Wnt通路信号转导变化比较复杂,其中WNT4、WNT8B、DKK2、CTNNB1改变超过2倍。Real-time PCR验证芯片结果可靠。[结论]具干细胞特性的食管癌放射抵抗细胞株与其亲本基因表达水平有明显差异。
[Purpose] To compare differential gene expression profiles between radioresistant esophageal carcinoma cells which acquired the CSC phenotype and parental cell line. [Methods] The two different groups were analyzed with Agilent feature extraction; the differentially expressed genes were selected and GO analysis was performed as well as pathway analysis. Real-time PCR was performed to confirm the microarray results. [Results] Two thousand two hundred and forty-six genes (fold change cut-off≥ 2) were differently expressed between two groups. Gene ontology analysis showed that 75.11 percent differentially expressed genes grouped to signal transducer activity within molecular function. In biological process, 35.94 and 25.8 percent differentially expressed genes related to cellular process and biological regulation. In cellular component, cell part and extracelluar region changed. The expressions of GLI2, MMP3, MMP9 were upregulated, PTCH1, FOXL1, BMP4, TGFB2, TGFB3 genes were downregulated. These results indicated that Hedgehog pathway might be upregulated and BMP-TGFβ downregnlated. Notch signal pathway might be downregnlated and Wnt signal pathway-related genes were complicated (WNT4, WNT8B, DKK2, CTNNB1 fold change cut-off 〉2). The results of real-time PCR and microarrays showed no significant difference. [Conclusion ] The patters of gene expression in the radioresistant esophageal carcinoma cells which acquired the CSC phenotype and parental cell line are obviously.
