摘要
从富油土壤中分离出9株产脂肪酶杆菌,其中BD3菌株产脂肪酶能力较强,通过测定其生理生化特征,初步鉴定该菌为假单胞菌属。利用聚乙烯醇(PVA)和硼酸交联作为固定化BD3菌株细胞的载体,并进行了转酯化的初步研究,发现由低浓度PVA制成的固定化细胞首次催化效率明显高于由高浓度PVA制成的固定化细胞,而且固定化细胞的酶活力与未固定化细胞的酶活力相比有所下降,但固定化细胞重复使用多批次后仍然具有活性。
9 strains producing lipase were isolated from soil with rich oil. Among them, strain BD3 was more powerful in lipase producing. Strain BD3 was identified as Pseudomonas sp according to its morphology, physiological and biochemical properties. We utilized PVA(polyvinyl alcohol) and Boric Acid as the Vector of Immobilized cells of Strain BD3, and studied transesterification at the same time. We found that catalytic efficiency of Immobilized ceils which was made of lower concentration PVA is higher than the high concentration PVA obviously. Besides that, the activity of immobilized cell will be weaker than the non-immobilized one. Immobilized cells maintained activity after many batches.
