摘要
目的优化白茅根多糖的提取工艺并对其含量进行测定。方法以白茅根多糖得膏率为考察指标,采用正交试验法对提取过程中的温度、次数、时间及料液比四因素进行优选研究,采用苯酚-浓硫酸法测定多糖含量。结果提取次数对多糖提取的影响最显著,其次是提取温度,提取时间和用水量对其影响较小。结论白茅根多糖的最佳提取工艺条件是用15倍水量在85℃温度下回流提取3次,每次水提取时间为3h。两产地的多糖含量均在0.15%以上。
Objective To optimize the extracting conditions of polysaccharide from Imperata Cylindrica and establish a suitable method for its content determination. Methods Based on the qualitative yield of polysaccharide extraction, a orthogonal test was employed to evaluate the effect of four factors including extracting temperature, extracting time, the extraction times and the ratio of material to liquid. The phenol-dense sulphuric acid method was applied for the content determination of polysaccharide. Results The times of extraction and the extracting temperature influenced the content of polysaceharides mostly, while the duration of extraction and the amount of water showed little influence. Conclusions The optimum extraction condition was refluxing and extracting for 3 times at 85 ℃ for 3 hours, and with 15 folds of water for every time. The contents of polysaccharide in Imperata Cylindrica collected from two different places were above 0.15%.
