摘要
目的:探讨银杏内酯B衍生物甲磺酸胺银杏内酯B(XQ)抗血小板黏附和释放作用。方法:通过血小板计数,测定XQ对胶原与家兔血小板接触引起的黏附的抑制作用。采用荧光分光光度法测定XQ对PAF诱导家兔血小板释放5-HT的影响,采用ELISA法测定XQ对PAF诱导家兔血小板释放β-TG、PF4的作用。结果:XQ1.95、3.9、7.8mg/kg对胶原与家兔血小板接触引起的黏附有抑制作用;XQ3.9、7.8mg/kg能显著抑制PAF诱导家兔血小板释放5-HT;XQ3个剂量组对PAF诱导家兔血小板释放β-TG和PF4有抑制作用。结论:XQ能够抗血小板黏附,减少血小板释放产物的生成,鉴于XQ的水溶性以及对血小板的作用,XQ治疗血栓性疾病可能比GB具有更大的优势。
Objective:To study antagonistic effect and mechanism of a new derivative of ginkolide B named XQ on platelet adhesion and release function.Method:With platelet count,to assay the effect of XQ on COL-induced platelet adhesion in rabbits,administrated at different final concentration via iv 5 days.With fluorospectrophotometry,to assay the effect of XQ on the release of 5-HT from PAF-induced platelet in rabbits.With ELISA,to assay the effect of XQ on the release of β-TG and PF4.Result:Three groups(1.95,3.9,7.8 mg/kg) have significant effect on inhibiting COL-induced platelet adhesion in rabbits,the inhibition ratio being 34.7%,55.8%,71.1% respectively(compared to normal P0.01).Two groups,3.9,7.8 mg/kg XQ,have significant effect on the release of 5-HT and from PAF-induced platelet in rabbits(P0.01).Three groups have significant effect on the release of β-TG and PF4(P0.05,0.01).Conclusion:XQ have the effect on inhibiting platelet aggregation and release function.Compared to GB,XQ may have more advantage of curing thromboembolus diseases because of its water-solubility and the function of inhibiting platelet aggregation and release.
