摘要
目的探讨姜黄素(curcumin,Cur)对人癌细胞株的体外作用特点及其水溶液的稳定性。方法以MTT法观察对人胃癌MGC803、人肝癌Be17402、人红白血病K562及其耐阿霉素株K562/ADM等的体外杀伤作用,并以体外活性变化为指标观察Cur水溶液的稳定性。结果Cur8.5~136.0μmol·L-1对上述细胞的IC50分别为13.0,15.9,11.7,62.6μmol·L-1;低于8.5μmol·L-1就能完全对抗41.5nmol·L-1表皮生长因子(EGF)的促增殖作用。Cur水溶液于4℃贮存3~7d体外抗癌活性明显下降。结论Cur对MGC803、Be17402、K562等有明显的杀伤作用,并能对抗EGF的促增殖作用,其水溶液稳定性较差。
AIM To investigate antitumor effects of curcumin (Cur) on human tumor cell lines in vitro and the stability of Cur in water solution. METHODS The cytotoxic effects of Cur on MGC803 .Be17402. K562 and K562/ADM cell lines were determined with MTT method; morphological change were observed with light microscope; the stability of Cur in water solution was determined by observing its activity of cytotoxicity in vitro. RESULTS IC50 of Cur on these cell lines exposed to the drug for 48 h were 13. 0, 15. 9, 11. 7, 62. 6 μmol· L-1 respectively; Cur at the concentration lower than 8. 5 μmol· L-1 was able to reverse the proliferative effect of EGF 41. 5 nmol· L-1 on MGC803 cells completely. When water solution of Cur at PH 7. 8 was stored at 4℃ for 3 days to 7 days, its antitumor activity in vitro decreased remarkely. CONCLUSION Cur showed potental cytotoxicity on human tumor cell lines and its water solution was relatively unstable.
