摘要
设计并合成一对特异性引物,从ConA激活的卢氏鸡淋巴细胞中扩增IL-21全基因并进行序列测定,应用多种生物学软件对卢氏鸡IL-21基因及其编码蛋白进行分析和三维结构预测。从具有中国地方特色的卢氏鸡中克隆了IL-21全基因。该基因全长438bp,编码145个氨基酸,包括19个氨基酸长度的信号肽,基因位于鸡的4号染色体55218k~55224k之间;编码的蛋白质分子量约16.67kD,为碱性蛋白,具有亲水性,无二硫键。三维结构预测表明:卢氏鸡IL-21蛋白主要由四个α螺旋和不规则卷曲构成,与人IL-21蛋白三维结构相似。同源性分析表明:该序列与其他哺乳动物IL-21的氨基酸相似性低于35%。
A pair of primers were designed for cloning IL-21 gene from total RNA of peripheral blood lymphocytes of Lushi chicken stimulated by ConA via RT-PCR technology.Lushi chicken IL-21 gene and the protein were analyzed by a variety of biologica software.The molecular model of three-dimensional structure of the IL-21 protein was predicted.The results revealed that the IL-21 gene of Lushi chicken is consistently 438bp in length,encoding for a predicted polypeptide of 145 amino acid residues,including a signal peptide with 19 amino acid residues.The gene is located on chromosome 4 between 55 218 k and 55 224 k.The gene encodes a 16.67 kD protein which is a hydrophilic basic protein.The predicted tertiary structure of the protein shows that Lushi chicken IL-21 protein contains random coil and four α-helix.This structure is similar with human IL-21 protein.Homologies of IL-21 amino acid sequence between Lushi chicken and another mammalian are all less than 35%.
