摘要
以磁性壳聚糖微球为载体,采用吸附-交联法固定化糖化酶。对给酶量、戊二醛添加量、交联温度、交联时间等因素进行了考察;并对固定化酶的酶学性质进行了研究。结果表明,固定化的最佳条件为:浓度为0.5 mg/mL酶液8 mL、1%戊二醛溶液4 mL、交联温度45℃、交联时间4 h;固定化酶的热稳定性和pH稳定性都优于游离酶;固定化酶连续使用5次,酶活仍保持了58%。
Saccharifying enzyme was immobilized by absorption-cross-linking method with magnetic chitosan microspheres as carriers. The influ- encing factors including enzyme concentration, the addition level of glutaraldehyde, cross-linking temperature, and cross-linking time etc. were in- vestigated and the enzymatic properties of the immobilized enzyme were studied. The results showed that the optimum immobilization conditions were as follows: 8 mL enzyme solution (its concentration was 0.5 mg/mL) used, addition of 4 mL 1% glutaraldehyde solution, cross linking temperature was at 45 ~C, and cross-linking time was 4 h. The thermal stability and pH stability of the immobilized enzyme was better than that of free enzyme and 58 % enzyme activity preserved even after continuous use of immobilized enzyme for five times
