摘要
目的探讨Rho/Rock信号通路在原发性高血压肾纤维化中的作用。方法选择31例原发性高血压病人,依据24h尿微量清蛋白排泄率(UAER)分为正常清蛋白尿组(A组)11例、微量清蛋白尿组(B组)10例和尿毒症组(C组)10例,选取正常体检者10例作为对照组,用酶联免疫吸附法(ELISA法)测定各组血清转化生长因子β(TGF-β)、基质金属蛋白酶9(MMP-9)及Rho关联含卷曲螺旋蛋白激酶1(ROCK-1)水平,分析TGF-β、MMP-9、ROCK-1在各组间的表达规律及三者之间的相关性。结果与对照组比较,A、B、C组TGF-β、MMP-9、ROCK-1血清浓度均显著升高(F=14.498~126.037,q=3.222~20.617;P<0.05、0.01);B、C组TGF-β及ROCK-1血清浓度较A组显著升高(q=4.797~17.636,P<0.01),B、C组间TGF-β、ROCK-1血清浓度差异无显著(q=0.612、1.175,P>0.05);B组MMP-9血清浓度较A、C组显著增高(q=4.799、5.959,P<0.01),而A、C组之间差异无显著性(q=1.299,P>0.05)。血清TGF-β与MMP-9、TGF-β与ROCK-1、MMP-9与ROCK-1浓度之间均呈正相关(r=0.381~0.652,P<0.05)。结论Rho/Rock信号通路可能参与了高血压肾纤维化过程。
Objective To investigate the effect of Rho/Rock signaling pathway on renal fibrosis in primary hypertension. Methods Thirty-one patients with primary hypertension were divided into normal-proteinuria group (group A, 11 cases), mieroalbuminuria group (group B, 10) and uremic group (group C, 10) based on urinary albumin excretion rate in 24 hours. Ten healthy adults served as normal control group (group NC). Enzyme-linked immunosorbent assay was performed to determine the serum concentrations of TGF-β,MMP-9 and ROCK-1. Expression patterns of TGF-β,MMP-9 and ROCK-1 and the correlation among the serum concentrations of TGF-β,MMP-9 and ROCK-1 were analyzed by ANOVA and S-N-K test. Results The serum TGF-β, MMP-9 and ROCK-1 in groups A, B and C were significantly increased than that of the control group (F= 14. 498- 126. 037,q= 3. 222--20. 617;P〈0.05,0.01). The serum TGF-β and ROCK-1 in groups B and C were significantly increased than that of group A (q:4. 797-17. 636;P〈0.01), but the difference of that between groups B and C was not significant (q=0. 612, 1.175;P〉0.05). The serum MMP-9 of group B was significantly increased than that of groups A and C (q= 4. 799,5. 959 P〈0.01), but the difference of that between groups A and C was not significant (q=1. 299, P〉0.05). There existed significant correlation between TGF-β and ROCK-1, MMP-9 and ROCK-1, MMP-9 and ROCK-1 (r= 0. 381- 0. 652, P〈0. 05). Conclusion It is likely that Rho/Roek signaling pathway involves in the process of renal fibrosis in primary hypertension.
