摘要
目的观察新生大鼠坏死性小肠结肠炎(necrotizing enteroeolitis,NEC)中c—Jun氨基末端激酶(c-JunN—terminalKinase,JNK)的激化及胞内分布规律,探讨其在NEC发病机制中的作用。方法新生鼠随机分为正常对照组和NEC动物模型组,每组10只,于母鼠喂养3d,第4d处死。取回盲部肠组织1~2cm,用10%的甲醛立即固定,常规石蜡包埋,分别作组织形态学检查和免疫组织化学检测JNK的表达。结果新生大鼠NEC模型中JNK在肠道细胞的胞浆中黄棕色染色颗粒表达并伴有向核转移,其阳性表达明显高于正常对照组。结论JNK信号通路可能参与了NEC发病过程,并起着信号传导作用。
Objective To investigate the intracellular distribution and role of c-Jun N-terminal Kinase (JNK) in hypoxia-induced neonatal rat necrotizing enterocolitis (NEC). Methods Newborn Wistar rats were divided randomly into experimental group and control group, 10 rats in each group. After NEC model was es- tablished, they were returned to their mothers. Rats were killed on day 4, and distal ileum was harvested for morphological study and immunohistochemistry for JNK. Results In normal intestinal tissue, there is no or only very weak staining for phosphorylation of JNK protein. Compared with control group, the expression of JNK was significantly increased and began to translocate from cytoplasm to nucleus in NEC group. Conclusion JNK signal pathway might play important role in the pathogenesis of NEC.
