抗氧化作用在锌干预大鼠胚胎心脏畸形发生中的意义 被引量:3

摘要 目的:探讨锌干预同型半胱氨酸(Homocysteine,HCY)诱发大鼠胚胎心脏发育畸形中抗氧化应激的意义。方法:SD成年健康清洁级雌雄大鼠各40只,常规方法交配获取孕鼠后,随机分为5个组:①空白对照组,②HCY组,③补锌组,④叶酸组,⑤联合组。各组孕鼠分别于妊娠第13、15、17、19天剖腹取出胚胎,通过切片进行胎鼠心脏组织学畸形观察。取胚胎时抽取孕鼠静脉血检测血清CuZn-SOD活力值及RT-PCR法半定量测定孕鼠外周血淋巴细胞金属硫蛋白-1型基因mRNA表达。结果:HCY组与空白对照组相比,吸收胎死胎率、心脏畸形率及不良胚胎率均明显增高(χ2=15.70、35.37、56.53,P<0.001);主要畸形表现为发育延迟、心房缺如、室间隔缺损、心室缺损及大体畸形等,补锌组与HCY组比较,吸收胎死胎率、心脏畸形率及不良胚胎率均明显减少(χ2=4.45、15.81、22.85,P<0.05、0.001、0.001);干预组间两两比较各率无明显改变(各组P>0.05);HCY组孕鼠血清CuZn-SOD活力值较对照组明显下降(t=8.28,P<0.005)、外周血淋巴细胞MT-1 mRNA表达较对照组明显下降(P<0.001);补锌组与HCY组比较CuZn-SOD活力值则明显增加(t=5.54,P<0.01)、MT-1基因表达亦明显增加(P<0.001)。结论:补锌组孕鼠CuZn-SOD活力值及MT-1 mRNA表达明显增加,不良胚胎率和胚胎心脏畸形率明显下降,表明锌元素能拮抗HCY对大鼠胚胎心脏的致畸作用,其机制与孕鼠抗氧化应激能力增强有关。 Objective:To explore the antioxidant effect of zinc on heart defects in rat embryos induced by homocysteine(HCY).Methods:40 SD-healthy adult male rats and 40 SD-healthy adult female rats copulated to get pregnant rats,then they were divided into control group,HCY group,zinc group,folic acid group and joint group.All the embryos were removed on the thirteenth,fifteenth,seventeenth and nineteenth day to observe fetal heart malformation through slicing,and serum Cu-Zinc SOD activity value was detected,the expression of peripheral blood lymphocytes MT-1 mRNA was detected by RT-PCR semi-quantitative method.Results:The rates of resorption and death,heart deformity and vicious fetuses in HCY group were significantly higher than those in control group(χ2=15.70,35.37,56.53,P0.001,respectively),the main malformation types included developmental delay,atrial absence,ventricular septal defect,ventricular defect and whole malformation.The rates of resorption and death,heart deformity and vicious fetuses in zinc group were significantly lower than those in HCY group(χ2=4.45,15.81,22.85,P0.05,0.001,0.001);there was no significant difference among subgroups in intervention group(P0.05);the serum Cu-Zinc SOD activity value and the expression level of peripheral blood lymphocytes MT-1 mRNA in HCY group were significantly lower than those in control group(t=8.28,P0.005)(P0.001);the serum Cu-Zinc SOD activity value in zinc group was significantly higher than that in HCY group(t=5.54,P0.01),the expression level of peripheral blood lymphocytes MT-1 mRNA in zinc group was significantly higher than that role in HCY group(P0.001).Conclusion:The serum Cu-Zinc SOD activity value and expression level of peripheral blood lymphocytes MT-1 mRNA of pregnant rats in zinc group increase,and the rats of vicious fetuses and heart deformity decrease significantly,the results reveal that zinc plays an antagonistic role in heart deformity induced by HCY,the mechanisms are related to strengthened anti-oxidative stress ability.
