黄粉虫壳中的壳聚糖提取工艺研究

摘要 以黄粉虫在生长过程中的皮蜕为原料,对提取壳聚糖的方法及工艺进行初步探索。结合他人经验,分析了最节约成本的脱除灰分等杂质的方法并且利用试剂浓度、温度和反应时间等单因素实验,系统研究了不同工艺条件对壳聚糖的脱乙酰度的影响,以期为工业生产提供依据。最终得到最佳提取工艺为:先将原料在室温下盐酸溶液浓度为1mol/L搅拌反应3.5h,再于沸水浴下与10%氢氧化钠溶液搅拌反应5h,接着用6%双氧水室温搅拌4h进行脱色处理,最后150℃油浴,氢氧化钠溶液浓度为50%,反应时间为8h,采用两段法处理得到壳聚糖。 With the caparace of Tenebrio molitor in growing process as starting material, the extration technology of chitosan was studied.Combining with the experience of others, how to remove ash and protein was analysed by single factor experiment such as the reagent concentration, temperature and reaction time. Also, systematically analyzed the effect of different technology condition on degree of deacetylation. The optimum conditions were obtained. The ultimate best condition is that using 1.5 mol/L hydrochloric acid with stirring the material for 3.5 hour, and then within NaOH when it's concentration is 10% for reacting 5 hour. Thirdly with the stuff stirring in H202 when it's concentration is 6% for 4 hour in room temperature. In the end, within the 50% NaOH for reacting 8 hour in interval, we can finally get chitosan.
