摘要
目的研究三氧化二砷(arsenic trioxide,As2O3)和四乙铵(tetraethylammonium,TEA)对人肺腺癌细胞AG-ZY-83-a增殖和凋亡的影响,探讨As2O3诱导人肺腺癌细胞株凋亡的机制。方法体外细胞培养人肺腺癌AGZY-83-a细胞株,采用MTT法检测As2O3和TEA对细胞增殖的影响。透射电镜(TEM)、流式细胞术(FCM)检测细胞凋亡情况。比色法检测细胞内Caspase-3活性改变。结果As2O3和TEA对AGZY-83-a细胞增殖有明显的抑制作用(P<0.01)。As2O3组对AGZY-83-a细胞增殖抑制率低于TEA+As2O3组(P<0.05)。As2O3和TEA使AGZY-83-a细胞凋亡率,Caspase-3增加,单用As2O3组的细胞凋亡率、Caspase-3活性分别高于TEA+As2O3组(P<0.05)。结论应用TEA部分抑制了As2O3诱导AGZY-83-a细胞凋亡;As2O3诱导AGZY-83-a细胞凋亡可能和As2O3对细胞膜不同的钾离子通道作用有关系。
Objective To investigate the effect of arsenic trioxide(As2O3) and tetraethylammonium on proliferation and apoptosis of human pulmonary adenocarcinoma AGZY-83-a cell line and its mechanism.Methods The inhibitory rate of cultured AGZY-83-a cells treated with As2O3,TEA was observed by MTT assay.Apoptosis was detected by flow cytometry and electronic microscope.Cellular caspase-3 activities were observed with colorimetric method.Results The proliferation of AGZY-83-a cells was inhibited by As2O3 and TEA(P0.01);Inhibitory rate in group treated with As2O3 was lower than that treated with TEA+As2O3(P0.05).As2O3 and TEA increased the percentage of apoptosis and the caspase-3 activity.The percentage of apoptosis and caspase-3 activity were higher in As2O3-treated group(P0.01) compared with those in TEA+As2O3-treated group.Conclusion The apoptosis percentage of AGZY-83-a cells induced by As2O3 is reduced partially by TEA;The mechanism of As2O3-induced apoptosis of AGZY-83-a cells is possibly due to the influence of As2O3 on multiple potassium channels in the cellular membrane.
