摘要
目的建立变应性鼻炎(allergic rhinitis,AR)小鼠模型,研究金黄色葡萄球菌肠毒素B(SEB)和卵清蛋白(OVA)在建立AR模型中的作用。方法将40只6~8周Balb/c小鼠随机分为4组:OVA组、SEB组、OVA+SEB组、生理盐水组,并建立小鼠AR模型。采用析因设计分析各组小鼠临床症状评分、血清IgE和IL-4的水平,并对鼻黏膜行组织形态学观察。结果OVA组、SEB组、OVA+SEB组及生理盐水组小鼠的症状评分分别为:0.90±0.99、0.70±0.82、6.80±1.03、0.60±0.70.OVA、SEB交互效应P〈0.01,有统计学意义,且OVA+SEB组鼻黏膜出现明显的AR形态学改变。血清IgE、水平分别为25.41±3.87ng/ml、20.11±2.23ng/ml、122.72±5.68ng/ml、18.67±2.00ng/ml;IL-4的水平分别为27.37±1.52pg/ml、24.05±2.13pg/ml、63.73±3.81pg/ml、23.50±2.62pg/ml,二者OVA、SEB交互效应均有统计学意义。结论只有SEB和OVA协同作用后可致小鼠AR,即SEB可以提高机体对OVA的易感性,发挥免疫佐剂的作用。
Objective To establish mice modle of allergic rhinitis(AR) and to study the role of Staphylococcal enterotoxin B (SEB) and ovalbumin(OVA) in the modle. Methods Forty Balb/c mice were evenly randomized into OVA group , SEB group, OVA + SEB group and normal sodium group and AR modle was established. The symptom scores,total serum IgE concentration, IL- 4 concentration were analyzed by factorial design. Meanwhile, the morphology change of nasal mucosa was observed. Results The symptom scores in OVA group, SEB group, OVA + SEB group and normal sodium group were 6.80 ± 1.03,0.90 ±0.99,0.70 ± 0.82,0.60 ± 0.70 respec-tirely. The interaction of OVA and SEB had statistical significance(P 〈0. 01 ). Nasal mucosa of OVA + SEB group displayed an AR morphology change . Meanwhile, Total serum IgE concentrations were 25.41 ± 3. 87μg/ml, 20.11 ± 2.23 μg/ml, 122.72 ± 5.68 μg/ml, 18.67 ± 2. 00μg/ml respectirely. IL- 4 concentration were 27.37 ±1.52pg/m1,24.05 ±2. 13pg/ml, 63.73 ± 3.81pg/ml,23.50 ± 2.62pg/ml respectirely, the interaction of OVA and SEB in IgE and IL - 4 concentration had statistical significance too. Conclusion Only OVA and SEB cooperation could induce AR development. SEB can elevate the affectivity of OVA to the organism and play a role of immunity adjuvant.
