摘要
目的观察旋毛虫免疫核糖核酸(iRNA)对BALB/c小鼠体内SP2/0肿瘤细胞的抑制作用。方法以不同剂量旋毛虫肌幼虫可溶性抗原免疫ICR小鼠、家兔和山羊,ELISA检测抗体效价达1:1024的最低免疫剂量确定为最佳免疫剂量,同时取各实验动物肝、脾和淋巴结,制备旋毛虫iRNA,并进行鉴定。以不同剂量iRNA接种BALB/c小鼠,并在不同时间接种SP2/0肿瘤细胞(试验分为先荷瘤和后荷瘤进行)。在荷瘤20d时,处死小鼠,测量肿瘤体积和重量,并检测脾脏T淋巴细胞亚群的变化。结果旋毛虫肌幼虫可溶性抗原的最佳免疫剂量分别为:ICR小鼠:300μg/只;家兔:2mg/只;山羊:20mg/只。制备的旋毛虫iRNA各项指标均合格。各试验组小鼠肿瘤体积和重量均小于相应的对照组,且差异均有统计学意义;各试验组小鼠的CD3+、CD4+及CD4+/CD8+比值较相应的对照组均有不同程度的增高。在后荷瘤试验组中,1mg组抑瘤效果最好;在先荷瘤试验组中,4mg组抑瘤效果最好。结论旋毛虫iRNA对BALB/c小鼠体内SP2/0肿瘤细胞的生长具有较强的抑制作用。
Objective To observe the inhibitory effect of immune RNA(iRNA)of trichinella on the SP2/0 tumor cells in BALB/c mice.Methods ICR mice,rabbits and goats were immunized with trichinella muscle larva crude antigen at various dosages separately,and the minimum dosage inducing an ELISA titer of 1:1 024 was served as the dosage for immunization.The livers,spleens and lymph nodes of the animals were isolated,from which iRNAs were prepared and identified.BALB/c mice were immunized with the prepared iRNAs at various dosages,and transplanted with SP2/0 tumor cells at various time points before and after immunization respectively.All the mice were killed 20 d after transplantation with SP2/0 tumor cells,and the size and weight of tumors were measured,while the T lymphocyte subsets in spleens were determined.Results The optimal dosages of trichinella muscle larva crude antigen for immunization of each ICR mouse,rabbit and goat were 300μg,2 mg and 20μg respectively.All the indexes of prepared trichinella iRNA met the relevant requirements.Compared with those in the corresponding control groups,the size and weight of tumors of mice in test groups decreased,while CD3+and CD4+lymphocytes as well as ratio of CD4+to CD8+lymphocytes increased at different degrees.The optimal dosages of iRNA for inhibiting the tumors in mice transplanted with SP2/0 cells before and after immunization were 4 and 1 mg respectively.Conclusion Trichinella iRNA showed strong inhibiting effect on the growth of SP2/0 tumor cells in BALB/c mice.
