摘要
【目的】采用双波长法同时测定谷类和豆类作物籽粒中直链和支链淀粉的含量。【方法】由直链淀粉一碘、支链淀粉一碘复合物的吸收光谱,确定直链和支链淀粉的波长对分别为495、565、530和654nm,将其代入回归方程,可求出样品中直链和支链淀粉的含量。【结果】直链淀粉的浓度在0~80mg/g、支链淀粉的浓度在0~400mg/L有很好的线性关系,即符合比耳定律。【结论】双波长法可以有效地消除吸收池和试剂等因素引起的误差,具有准确度高等特点,用该方法测定直链和支链淀粉的含量得到了很好的结果。
[ Objective ] The paper mainly uses dual - wave length spectrophotometry method to study the determinnation of amylose and amylopectin in grain and bean. [Method]Based on the absorption spectra of the two complexes,the amylose- wave length of 495/565 nm,and amylopectin wave length of 530/654 nm were obtained as the measuring wave length. According to the linear regression equationes, the paper can determine the content of amylose and amylopectin in sample solution. [ Result] The result shows that amylose concentration is between 0 - 80 mg/L and amylopectin concentration is between 0 - 400 mg/L. [ Conclusion ] The content of amylose and amylopeetin determined by the method is accurate, because it is not influenced by the reagent and the error which is caused by the absorption cell . So that it is most suitable to determine amyloy and amylopeetin and the results of the experiments is most satisfactory.
