摘要
目的:评价17997(20mg·L-1)滴眼治疗实验性单纯疱疹性角膜炎(herpessimplexkeratitis,HSK)的疗效及滴眼剂的刺激性。方法:建立兔眼HSK动物模型,以1g·L-1无环鸟苷滴眼剂作为阳性对照,以0.15mol·L-1生理盐水作为阴性对照,采用裂隙灯观察角膜病变情况并评分、病毒分离检测及透射电镜观察评价17997疗效。采用Draize眼刺激实验评价药物毒性。结果:17997(20mg·L-1)可有效地治疗HSK,减轻角膜病变程度,缩短平均治愈时间,疗效与无环鸟苷相似。17997滴眼剂(20mg·L-1)及其赋形剂的Draize评分结果表明,17997滴眼剂无眼刺激性。结论:广谱抗病毒药17997治疗实验性单纯疱疹性角膜炎获得了与无环鸟苷相似的疗效。
Objective: To evaluate the efficacy and toxicity of 17997 against experimental herpes simplex keratitis(HSK) in rabbits. Methods: Rabbit eye model of HSK was constructed. 1 g·L -1 acyclovir (ACV) and normal saline were used as positive and negative control, respectively. The antiviral efficacy of 17997was evaluated by slit lamp observation, virus isolation and transmission electron microscope observation. Draize irritation test was applied to evaluate the eye toxicity of 17997 in vivo . Results: Compared with normal saline group, both topical 17997 (20 mg·L -1 ) and 1 g·L -1 ACV significantly reduced severity of HSV Ⅰ dendritic keratitis, shortened healing time of HSK. There was no statistical significance between 17997 and ACV. The result of Draize test showed that 17997 and its solvent had no toxicity. Conclusion: The efficacy of 17997 against HSK was similar to that of ACV and there was no clinical toxicity.
