摘要
目的:探索静态瞬稳注射化学发光法检测大豆异黄酮方法。方法:本实验在氢氧化钠-铁氰化钾体系下对大豆异黄酮进行化学发光分析,确定较佳实验条件,利用光强度测定大豆异黄酮含量。结果:获得理想发光曲线的实验条件是在-1000 V负高压、30 s发光时间,当铁氰化钾溶液浓度为0.16 mol/L,氢氧化钠溶液浓度为0.04 mol/L时,整个反应体系的化学发光最强。大豆异黄酮的浓度在0~200.00 mg/L范围内,与化学发光强度成良好线性关系,相关系数r=0.997。结论:此方法简单,周期短,成本低。加标回收率95%~110%,RSD5%(n=3),检测限为0.1 mg/L,测试结果较理想,能满足大豆异黄酮产品质量检测的需要。
Objective:To explore a new method for determiniation of soybean isoflavones by static instantaneous steady injection chemiluminescence.Methods:In this experiment,soybean isoflavones was detected and analyzed by using chemical luminescence with high sensitivity in the sodium hydroxide-potassium ferricyanide system,and the best experimental conditions was found out.Results:The luminescence curve of the most idea experimental conditions was negative high voltage-1000 V,light-emitting time of 30 s.And the concentration of potassium ferricyanide solution was 0.16 mol/L,sodium hydroxide was 0.04 mol/L.Chemiluminescence of the entire reaction system was the strongest.We could gain a good linearity with correlation coefficient 0.997 in the concentration of soybean isoflavones 0~200.00 mg/L.Conclusion:The method is simple,short-time and low-cost.The recoveries of standard addition is 95%~110%,and relative standard deviation lower than 5%(n=3),detection limit 0.1 mg/L.The test result is ideal.This new method should provide experimental basis and quality inspection needs for the detection of soybean isoflavones.
