Relationship between Toll-like receptor 4 protein and atherosclerosis

摘要 目的探讨Toll样受体4(TLR4)的表达与动脉粥样硬化(AS)及其主要危险因素的关系。方法选取行冠状动脉旁路移植术患者31例共63块主动脉壁全层组织作为研究组;肾移植供体者8名共32块动脉壁全层组织作为对照组。应用免疫组织化学方法检测动脉壁全层组织中TLR4蛋白的表达,并分析TLR4蛋白的表达与年龄、性别、吸烟、血压、血糖、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、总胆固醇(TC)、甘油三酯(TG)等AS危险因素之间的关系。结果TLR4在研究组动脉壁全层组织中有明显表达,而对照组无表达。TLR4表达与高血压、高血糖、LDL-C升高、HDL-C降低相关(P均<0.05),与年龄、性别、吸烟、TC、TG无相关性(P>0.05)。结论TLR4可能是AS的始动因素,高血压、高血糖、LDL-C升高、HDL-C降低等危险因素通过TLR4信号途径促进AS的发生发展。 Objective To investigate the changes of Toll-like receptor 4(TLR4) in the aortas of patients with coronary atherosclerosis and its association with atherosclerotic risk factors.Methods 63 pieces of aorta tissues from 31 patients undergoing coronary artery bypass graft(CABG) surgery were taken as the study group.Another 32 pieces of aorta tissues from 8 healthy young men through the kidney donation program were taken as control.Expression of TLR4 protein was determined by immunohistochemistry.The relationships of TLR4 protein expression and the risk factors of atherosclerosis,including age,gender,smoking,hypertension,hyperglycemia,high density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-C),total cholesterol(TC) and triglyceride(TG) were analyzed.Results TLR4 protein was detectable in aortas from CABG patients.However,it was absent in the control aorta tissues.Expression of TLR4 protein was correlated with hypertension,hyperglycemia,increased LDL-C and decreased HDL-C(P〈0.05),but not correlated with age,gender,smoking,TC and TG(P〉0.05).Conclusion Our study demonstrates that the TLR4 protein may be an initiator factor of atherosclerosis.The risk factors including hypertension,hyperglycemia,increased LDL-C and decreased HDL-C which work partly through the TLR4 signal pathway.
