不同干预治疗对C57BL/6J雄性肥胖小鼠肿瘤坏死因子-α的影响 被引量:6

摘要 目的探讨不同干预治疗对高脂饮食诱导的C57BL/6J雄性肥胖小鼠脂肪组织肿瘤坏死因子-α(TNF-α)的影响。方法4周龄C57BL/6J雄性小鼠随机分为普食组(以普通饲料喂养,脂肪占总热量的12%)、高脂组(以高脂饲料喂养,脂肪占总热量的12%)喂养,再将40只高脂饮食诱导的C57BL/6J雄性肥胖小鼠随机分为饮食控制组、高脂饮食组(高脂饮食)、二甲双胍组(高脂饮食+二甲双胍)及罗格列酮组(高脂饮食+罗格列酮)。干预治疗4周后,分别测定体重、内脏脂肪重量、空腹血糖(FBG),空腹胰岛素水平(FIns)。评价小鼠糖代谢和胰岛素抵抗情况,采用实时聚合酶链反应(RT-PCR)方法及Western印迹法检测脂肪组织内TNF-αmRNA、TNF-α的表达。结果①高脂组小鼠的体重、内脏脂肪重量、FBG、FIns水平和脂肪组织TNF-α、TNF-αmRNA的表达均显著高于普食组(P值分别<0.05、0.01)。②干预治疗1周后,饮食控制组、罗格列酮组的FBG、内脏脂肪重量和脂肪组织内TNF-α、TNF-αmRNA均显著低于高脂饮食组(P值分别均<0.05、0.01)。结论罗格列酮及饮食控制均能一定程度地降低肥胖小鼠的内脏脂肪重量、血糖,降低脂肪组织TNF-α的表达水平。 Objective To investigate the effect of different treatments on tumor necrosis factor-alpha expression in adipose tissues of high fat diet-induced obese mice.Methods The mouse obese model was established by high fat diet.Forty obese mice were randomly divided into 4 groups:diet control group(n=10,fed with normal diet containing 12% fat),high fat group(n=10,continued with high fat diet containing 60% fat),Metformin group(n=10,Metformin + high fat diet) and Rosiglitazone group(n=10,Rosiglitazone+high fat diet).After a 4-week intervention,the body weights,visceral fat(VF),fast glucose,fast insulin levels of each group were assayed.At the same time,the expression of TNF-α mRNA and protein in adipose tissue was examined by RT-PCR and Western blotting methods.Results ① The body weight,VF,fast glucose,fast insulin and expression of TNF-αin fat tissue of the high fat group were significantly higher than those of the normal diet group(P〈0.05,0.01).② The fast glucose,VF,and TNF-α expression in the fat tissue in the diet control group and Rosiglitazone group were significant lower than those in the high fat group,(P〈0.05,0.01).Conclusion Rosiglitazone and diet control can effectively lower VF,fast glucose and the level of TNF-α in fat tissue of obese mice.
