摘要 目的观察探讨桂枝芍药知母汤对RA大鼠滑膜细胞增殖、凋亡的影响及对细胞凋亡调控基因的影响,探讨该汤药的作用机制。方法利用醋酸溶解Ⅱ型胶原蛋白及弗氏完全佐剂混合,诱导大鼠佐剂性关节炎模型,以"桂枝芍药知母汤浓缩液"为干预药物,行体内、体外双重实验,观察滑膜细胞凋亡及其形态改变情况,应用SP法检测Fas抗原、Bcl-2蛋白、p53蛋白。结果体内、体外实验实验组的作用均优于对照组(P<0.05),两实验中实验组细胞凋亡增加,Bcl-2、p53表达下降,Fas表达上调,与对照组及正常组对比差异有统计学意义(P<0.05)。结论桂枝芍药知母汤浓缩液可明显减轻类风湿性关节炎大鼠滑膜增殖的病理改变,并可使体内、体外实验组RA大鼠Bcl-2、p53表达下降,Fas表达上调,为临床应用桂枝芍药知母汤治疗RA提供了实验依据。 Objective To observe the effective of Gui zhi shao yao zhi mu decoction on RA synovial cell proliferation,apoptosis and regulation of cell apoptosis genes in rats,and explore the mechanism of this Decoction on the disease.Methods Rats were prepared for adjuvant arthritis models induced b acetic acid soluble typeⅡcollagen and Freund's complete adjuvant,and "concentrate Guizhi-shaoyao-zhimu soup" as the intervention drug;using in vivo,in vitro double-blind experiments to observe the changes of synovial cell apoptosis and morphous,meanwhile,the Fas antigen,Bcl-2 protein and p53 protein were detect by the method of SP.Results The result of in vivo and vitro experiment,the role of the experimental group were better than the control group(P0.05);in both experiment,synovial cell apoptosis and Fas expression were increased in the experimental group,while the Bcl-2 and p53 expression were decreased,comparing with the control group and normal group there were statistically signifi cant(P0.05).Conclusion Guizhishaoyaozhimu soup concentrate can signifi cantly reduce the proliferation of rheumatoid arthritis synovial in rats;the experimental group in botn vivo and vitro experiment,synovial cell apoptosis and Fas expression were increased in the experimental group,while the Bcl-2 and p53 expression were decreased.So this study had provided experimental basis for clinical application of Guizhishaoyaozhimu decoction.
