摘要
目的建立了棉籽仁中棉酚的高效毛细管电泳检测方法。方法使用长度为58.5 cm、内径75μm的毛细管柱,分离电压20 kv,进样量0.5 pis×10 sec,分离温度25℃,缓冲盐溶液为0.01 mmol/L四硼酸钠-磷酸(pH=8.00)检测波长235 nm。结果棉酚在5.0-50.0μg/ml范围内线性良好,r=0.999 8,检测限为0.08μg/ml,加样回收率为97.5%-98.4%。新疆南疆不同品种棉籽仁中棉酚含量为0.60%-1.04%。结论本法具有良好的精密度和回收率,可作为棉籽仁中棉酚的含量测定方法。
Objective High performance capillary electrophoresis(HPCE) method was established for the separation of gossypol in cotton seed kernel.Methods The separation was carried out on a fused-silica uncoated capillary column (58.5 cm ×75 μm).The sample was injected by hydro-dynamic injection(0.5 Pis×10 sec) at a temperature of 25℃.The analysis was completed in 0.01M sodium tetraborate-phosphoric acid buffer(pH8.00) at 20 kV of separation voltage and 235 nm of detection wave length.Results The linear range of gossypol was 5.3-49.8 μg/ml and the correlation coefficient was 0.999 8.The limits of quantitation were 0.08 μg/ml The average recoveries were in the range of 97.5%-98.4%.Gossypol content of different cottonseed in Southern Xinjiang was 0.60%~1.04%.Conclusion The method has good precision and recovery.It can be used for determination of gossypol in cotton seed kernel.
