吡咯喹啉醌对大鼠雪旺细胞增殖及其EGR1、EGR2基因表达的影响 被引量:3

摘要 目的观察吡咯喹啉醌(PQQ)对大鼠雪旺细胞的促进增殖作用及其EGR1、EGR2基因表达的影响。方法体外原代培养大鼠雪旺细胞,纯化及S-100免疫荧光鉴定;加入PQQ进行培养,使PQQ终浓度分别为0、1、10、100、1 000、10 000 nmol/L,持续作用72 h,提取总RNA进行RT-PCR,检测EGR1及EGR2基因的表达变化。结果PQQ可促进雪旺细胞增殖并改变其形态,1~1 000 nmol/L的PQQ可使EGR1、EGR2基因表达增高,10 000 nmol/L时抑制EGR1及EGR2基因的表达。结论PQQ可促进大鼠雪旺细胞增殖并改变其形态,使雪旺细胞ERG1及EGR2基因表达上调。 Objective To investigate the effect of pyrroloquinoline quinine (PQQ) on proliferation and expression of EGR1 ,EGR2 on cultured Sehwann cells of rats. Methods Schwann cells of rats were cultured and purified in vitro and identified and purified by immunofluorescenee of S-100. Different concentration of PQQ (0,1,10,100,1 000,10 000 nmoL/ L) was added into culture medium for 72 h respectively. The expression of EGR1 and EGR2 gene were detected by RTPCR. Results Proper concentration of PQQ ( 1-1 000 nmol/L) could promote Sehwann cells proliferation and up-regulate the expression of EGR1, EGR2 on cultured Schwann cells ( P 〈 0.05). Conclusions PQQ may promote the proliferation, morphology and up-regulation of EGR1, EGR2 gene of rat Schwann cells.
