摘要
在纤维素-三(3,5-二甲基苯基氨基甲酸酯)(CDMPC)手性柱上,以正己烷-醇为流动相,对3种外消旋氨基酸衍生物(苯乙内酰脲)在正相模式下进行了拆分。考察了流动相中醇链长度、醇的立体结构、醇的含量对手性拆分的影响。首次用三元流动相体系对样品进行了拆分,发现利用三元流动相体系可达到最佳拆分效果。根据试验结果,对样品在CDMPC-CSP手性固定相上的正相手性识别机理进行了讨论。
Normal-phase chromatographic model was employed for enantiomeric separation of a-amino acid derivatives on the cellulose tris (3, 5 - dimethylphenylcarbamate - chiral stationary phase (CDMPC- CSP) ) in an eluant system consisting of a mixture of n-hexane and alcohol. The influence of alcohol such as the length of alkyl chain, steric bulk and content of alcohol in mobile phase were systematically examined. The result showed that the mobile phase strength and the structure of chiral compounds were dominant factors to retention and resolution of the analytes. Meanwhile, the steric effects of alcohol seemed to be a certain factor. We observed that when a ternary mobile phase was used, a good separation of enantiomers could be obtained. Based on the experimental results, the mechanism of the chiral discrimination on CDMPC-CSP column under nornal-phase chromatographic model was discussed.
