NS-398诱导尖锐湿疣患者外周血PBMC凋亡及对调控蛋白表达的影响

摘要 目的:评价环氧化酶2(COX-2)抑制剂NS-398对尖锐湿疣患者外周血单个核细胞(PBMC)凋亡及凋亡调控蛋白表达的影响。方法:采用流式细胞仪检测不同浓度NS-398作用CA患者PBMC24 h后细胞凋亡及凋亡调控蛋白的表达。结果:100-200μmol/L的NS-398对PBMC具有明显诱导凋亡的作用,随NS-398浓度增加,凋亡调控蛋白Apo-1、Apo-2.7表达水平呈下降趋势,Bcl-2表达水平呈上升趋势。结论:NS-398可以通过抑制COX-2的途径促进PBMC凋亡,使PBMC的凋亡调控失衡得以恢复。 Objective: To investigate the effect of cyclooxygenase - 2 inhibitor NS - 398 on the apoptosis and regulative protein expression of peripheral blood mononuclear cells (PBMCs) in condyloma acuminatum (CA). Methotis: Apoptosis and expression of regulative protein of PBMC cultured in different concentrations of NS - 398 medium for 24 h were detected by flow cytometry. Results: The apoptosis of PBMC cultured in 100 and 200 μmol/L NS - 398 medium for 24 h was increased. As the increase of concentrations of NS - 398, the expressions of regulative protein Ape - 1 and Ape - 2.7 were decreased. But the Bcl - 2 expression continued to increase. Conclusion: NS - 398 can induce apoptosis of PBMC and restore uncontroled apoptosis balance of PBMC.
