摘要
目的:研究超声技术提取葛根中葛根素的最佳条件,建立高效液相色谱法测定葛根中葛根素含量的方法。方法:采用超声技术提取葛根素,并利用正交试验法进行优选超声提取葛根素的最佳试验条件,采用高效液相色谱法,以葛根素为化学对照品,波长:250nm测定葛根中葛根素含量。结果:各因素对葛根素提取率的影响次序为:超声次数(超声时间(甲醇加入量(甲醇浓度。葛根素回归方程为:A=-152791.51+868038.61M,r=0.9997。精密度试验RSD%=1.0,仪器精密度良好;稳定性试验RSD%=1.3,在5h测定葛根素的含量是稳定的;重现性试验RSD%=2.39、平均回收率101.0%;结论:超声提取法提取效率高、操作简便、省时,是提取葛根有效成分的较好方法,值得推广。
Objective:To research the best method of extracting Puerarin from Pueraria.Establishing the method of determining the content of puerarin in Pueraria by HPLC.Methods:Extraction Puerarin by ultrasonic technology.The optimum extraction process by ultrasonic was the yielding contents of puerarin in extract which were determined by HPLC,puerarin for the standard,wavelength:250nm.Results:The influence order of each factor on the extracting of puerarin was that the extracting times﹥extracting time﹥methanol quantity﹥concentration of methanol.The return formula of puerarin is A=-152791.51+868038.61M,r=0.9997,Precision RSD=1.0%,equipment in good precision,Stability RSD=1.3%,determining the content of puerarin in 5 hours by HPLC is stable.repeatability RSD=2.39%,the average recovery rate of 101.0%.Conclusion:The ultrasonic extraction method is of high extracting efficiency,simple and time-saving.Which is a good method to extracte the active ingredients of Radix Puerariae lobatae,it is worth promoting.
